How the Analysis Works

Pre-column derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC), reversed-phase HPLC on a C18 column, and UV detection at 260 nm.

AQC reacts with primary and secondary amines in borate buffer to form stable urea derivatives that absorb at 260 nm. The excess reagent hydrolyzes to a by-product that elutes away from the amino acids. Cohen & Michaud, Anal. Biochem. 1993, 211, 279–287.

Why AQC chemistry

The derivatives are stable for days, secondary amines such as proline are tagged along with everything else, and the reaction runs to completion in ten minutes at 55 °C. It is the chemistry behind the most widely used commercial amino acid analysis kits.

How AQC derivatization works → · Methods library

  • An internal standard is added to every sample before derivatization to monitor the derivatization and injection.
  • A quality-control standard — an amino acid mixture of known concentration — is analyzed with every run to confirm the calibration holds.
  • Detector sensitivity is checked against the standard, and selected amino acids are spiked into samples where the matrix calls for a recovery check.
  • Reagent blanks are run alongside the samples; results below the quantitation limit are flagged rather than reported as numbers.

What we don't do. We measure free amino acids and ammonia only. We don't run acid hydrolysis, so we don't report total amino acid composition, protein content or peptide content.

Frequently Asked Questions

How does the pilot work?

New clients can send up to 5 samples for $250, one pilot per lab. You get the same report as any order, with the chromatogram and QC for each sample, and we are glad to talk through the results.

Do you measure total amino acids or protein content?

No. We measure free amino acids and ammonia, without hydrolysis. Total amino acid composition, protein content and peptide content need acid hydrolysis, which we do not offer.

What sample types can you analyze?

Liquid samples: fermentation broth, spent and defined media, culture supernatants, protein hydrolysates, wort, juice, beverages and aqueous extracts. Solids and powders are handled on request (PREP-1).

Which amino acids are reported?

The standard panel reports 19 proteinogenic amino acids and ammonia. The extended panel adds GABA, ornithine, citrulline, taurine and hydroxyproline. Cysteine / cystine is available on request.

How are results delivered?

As a PDF report with concentrations in µM and mg/L, the chromatogram and the run's quality-control summary. Raw data is available as CSV on request.

Is there a minimum number of samples?

No. One sample is a normal order; the minimum order value is $150. Volume pricing applies per order at 10 and 50 samples.

Is my sample information confidential?

Yes. Sample descriptions, results and client details are shared only with the submitting client.

Can you measure something other than amino acids?

Often, yes — through custom HPLC method development (MD-1). Start with the free feasibility call and we'll tell you whether the analyte and matrix are a good fit.
